如何测量肉眼看不见的特定区域

各位好，AFM初学者想问个问题：

基底上有一块5μmX5μm的区域，肉眼看不到，在实时画面中找到后，如何确保测量的是目标区域？把目标区域中心移到红色十字架处就可以吗？（x offset与y offset设为0是指红色十字中心？）
