关于用AFM测叶片表面形貌的问题

我想问问对于植物叶子样本应该怎么处理，我是直接将它用去离子水清洗干净，然后直接取样，用双面胶粘贴，但是过一段时间后，就粘不住了，不知道有没有什么好的处理方法。

还有，在扫描的过程中，高度图的信号时有时无，这是不是因为我叶子表面不规整的原因啊？谢谢了！
