Our faculty bought a new Catalyst version(8.15) and I am learning how to use the afm. I notice that there is a scanasyst mode and it does not need any tunning of the cantilever. Does it mean that I dont need to calibrate the spring constant and the deflection sensitivity? Can scanasyst help me get the foce-distance curve of everypoint on the cell?
Besides, I found the most recommended probe for scanning cell surface is DNP. Can I use DNP for scanasyst mode? Or I have to use the scanasyst( fluid, air) ? What spring constant of the cantilever should I choose? My note said I should use 18 tip half angle, what does it mean? is it a parameter that we need to adjust or it is talking about the tip itself? is there anything else that I need to pay attention to when scanning the cell? Thanks!!
