    Can you verify if you are using the relative trigger mode? Using the absolute trigger will likely give you different max applied force values depending on your deflection at the start of each force curve. Also, which probe are you using? I have not tried to apply such high forces much on Bio-samples but I wonder if it might be possible that your probe

    Hi Sarah, You can install the NanoScope Software on another computer (other than your AFM system). During installation, select the Nanoscope installation type as 'Offline Workstation'. Once installed you can email Bruker to get a key to enable MIRO in this offline version of the software. This will allow you to open .iro files and manipulate

    You should select the 'ramp and capture' option. Just 'ramp' only signals the AFM to perform the force measurements - it does not save the resulting force curves. With 'ramp and capture' the force curves will be saved in a folder with a file extension 'mca' (multi-curve analysis). If you open this folder in Nanoscope

    Hi Darice, The Sneddon model should be the better option for the modulus range of your samples. Unlike the DMT model, Sneddon does not use the tip radius so changing that shouldn't affect the modulus value in the Sneddon channel (you don't even need to calculate it).     Hi Benoit, Yes, you can use this probe for force volume measurements. Just be aware of how soft your sample is - as for samples like cells, bacteria and membranes this probe may be on the stiffer side (~0.7N/m). If you need a softer probe I've used the DNP-A probe (0.4N/m) which is about the same tip radius. Best, Andrea

    Hi Benoit, The deflection sensitivity of a probe does change depending on the particular AFM system you are using (different photodetectors) and depending on whether you are in fluid or air.
