Does anybody have any experience in that field? I would like to keep the bacteria ALIVE on the cantilever. In most publications the protocole is not detailed. So far, I tried with different concentrations of PolyLysine and PolyEthylene Imine on a tipless cantilever but the bacteria where always detaching after a few minutes of force pulling. Any trick would be much appreciated.

Thanks,

Alex.

Perhaps you are already doing it, but do the last washing step of the bacteria with demi water, suspend the centrifuged bacteria in 1 or 2 ml demiwater and use that while gluing the bacteria to the tip with poly L lisine. So while in demi and not in buffer. You have to do this quick, bacteria will not survive  long in demi.
