Hello Alex and Patrick,

Thanks for your answers. It's glad to hear that we set the scripts correctly.

However, I'm still doubtful about the plots under setpoint ramp mode. Is there any signal channel can be used to record the actual deflection, instead of the deflection error? If there is, then I could really see the deflection increases linearly at the pre-set ramp velocity.

And I'd also like consult you about our experimental setup. Please correct me if I'm wrong.

·         Firstly, since I'm approaching the bacterial probe to the inert surface, the "fetch molecule" step can be skipped. Am I right at this point?

·         And script 1 and 2 were supposed to do the same job: keep the cell under a constant force for some time. Hence, I would expect to see the similar plots from Height Sensor channel during the surface delay for both scripts.

·         Do I have any other option to achieve the same goal with the afm?
